Venetoclax is a BCL-2 inhibitor, a class of drugs that target the B-cell lymphoma 2 (BCL-2) protein. BCL-2 helps cancer cells survive by preventing programmed cell death (apoptosis). By inhibiting BCL-2, venetoclax promotes the death of cancerous cells. It is used to treat:

• Chronic lymphocytic leukemia (CLL)

• Small lymphocytic lymphoma (SLL)

• Acute myeloid leukemia (AML)

Venetoclax is typically administered in combination with other medications, such as rituximab, obinutuzumab, azacitidine, decitabine, or low-dose cytarabine, depending on the specific condition being treated. 

💊 Dosage Form

• Tablet: Venetoclax is available in oral tablet form. The typical starting dose for CLL/SLL is 20 mg daily, with a gradual increase over 5 weeks to a maintenance dose of 400 mg daily. For AML, the starting dose is 100 mg daily, escalating to 400 mg or 600 mg daily, depending on the combination therapy used. 

⚠️ Important Considerations

• Tumor Lysis Syndrome (TLS): Rapid breakdown of cancer cells can lead to TLS, a potentially life-threatening condition. Patients are monitored closely, and preventive measures are taken before starting treatment.

• Drug Interactions: Venetoclax is metabolized by the liver enzyme CYP3A4. Concomitant use with strong CYP3A4 inhibitors or inducers can affect venetoclax levels and should be managed carefully.
